Cristiano Ronaldo opened scoring from the penalty spot in the 13th minute.
Weston McKennie doubled the Serie A champions’ lead at the Nou Camp, before Ronaldo converted from 12 yards again.
The result takes Juve to the top of Group G, ahead of Barca.
This means Lionel Messi and colleagues will have to face a club that finished as group winners in the next round.
The list includes: Bayern Munich, Manchester City, Liverpool, Chelsea and Borussia Dortmund.
UEFA will conduct the draw on December 14.
